Phytase activity of phytase-producing bacteria isolated from mangrove sediment

Abstract Through the role of phytase, some soil bacteria can mineralize insoluble organic P such as phytate (myoinositol hexakisphosphate). Phytase is a secreted enzyme possessing ability to hydrolyze into phosphate esters and inorganic for plants absorption. This study aimed isolate phytase-producing from mangrove sediment examine influence nutrients (source carbon, nitrogen, phosphorus) physical conditions (temperature, pH, NaCl tolerance) on maximum phytase production. The presence activity was determined by examining individual colonies formation clear zone. Furthermore, isolates were screened qualitatively quantitatively using solid liquid screening medium (PSM) containing sodium Phytate substrates. result showed that total 48 have potential produce with production range 1.11 - 14.83U/mL. F15 Bacillus altitudinis found highest after 72 hours incubation, selected further analysis. strain resulted in optimal levels at 35°C pH 6.5 parameters, tolerated 5% lactose tryptone, which served carbon nitrogen sources, respectively.
